Green Fluorescent Protein

Green fluorescent protein (GFP) is a commonly used reporter construct throughout the field of biology. When excited by UV or blue light, GFP will fluoresce green, making it an excellent reporter construct1. GFP allows for the survival of the genetically modified plant unlike a GUS reporter, which is lethal to the plant2.
When observing the soybeans with the GFP reporter under UV light, green fluorescence was observed in both the transgenic soybeans and the control without transgenes. Because of this, a GFP reporter was not a good reporter to use in our soybean constructs. This is due to autofluorescence from excitement of molecules that are naturally occurring in plants by UV light. The most common of these molecules are chlorophyll and lignin3.
